The pathogenic role is virtually unknown, but CF patients infected with this fungus are at high risk of developing invasive disease. So far monitoring relies only on long-term fungal culture (4 weeks). Due to limitations of this procedure, the development of an additional easy-to-perform assay for detection of specific antibodies would facilitate monitoring. Therefore, an immunoassay was developed using commercially available precalibrated microaffinity columns (ABICAP TOOL, Biognosis, Jülich, Germany) containing ready-to-use activated solid phase matrix. By this means 108 sera from three groups (A: CF patients with and B: patients without E. dermatitidis recovered from sputum; C: healthy blood donors) were analyzed for prevailing IgG-, IgA- and IgE-antibodies using an antigen prepared from E. dermatitidis ex-type strain (CBS 207.35). In group A 83%, in group B 35% and in group C 6% of the test results for IgG, in group A 42%, in group B 24% and in group C 4% of the test results for IgA were above the upper limitting value. Thus, statistically significant differences between group A and C were revealed for IgG-, IgA and IgE-antibody production. In case of group A and B a significant difference was determined only for IgG-antibody production. In conclusion, the developed immunoassay proved to be well suited for serologic monitoring of colonized CF patients.","abstract_html":"Exophiala dermatitidis is a potentially pathogenic fungus frequently recovered from sputum of patients with cystic fibrosis (CF).
