The aim of the study was to investigate the pharmacodynamics of transdermal scopolamine (Scopoderm[R] TTS) under microgravity conditions and on earth. In microgravity, cardiovascular effects were analyzed by two astronauts/cosmonauts, on earth the tests were performed by four astronauts/cosmonauts and eight male volunteers. Under gravity conditions, transdermal scopolamine led to a significant decrease of heart rate at rest and in the recovery phase. Additionally the systolic, diastolic and mean blood pressure decreased significantly at rest, during cycle ergometer and in the recovery phase. Transdermal scopolamine did not influence the heart rate of the two astronauts/cosmonauts in microgravity. The systolic, diastolic and mean blood pressure was not systematicly affected by the drug. We conclude that the application of transdermal scopolamine has different effects on the cardiovascular system, depending on gravity. In microgravity, pharmacodynamics and pharmacokinetics might be influenced by alterations of the endocrine system, the cardiovascular regulation and the function of the kidney including regulation of body fluids and electrolytes.
