In this thesis, the overexpression of s-adenosylhomocystein hydrolase (SAH) was analyzed in EBV genome positive B95-8 cells. The focus of the examination has been on the signal transduction pathways which play a part in the activation of latent persistent EBV.The gene of SAH was brought in a fusion vector (pcDNA-AHCY) which was transfected in B95-8 cells by endocytosis. The successful transfection was proved by the evidence of ß lactamase on the vector pcDNA-AHCY.In pcDNA-AHCY transfected B95-8 cells an increased expression of the viral genes EA-D and BZLF1 was shown by immunofluorescence assay and PCR. The transfection with SAH led to an increase of phosphorylated and activated protein kinase C (PKC). The lytic cycle might be activated additionally using a PKC and PLA2 independent way.The rate of DNA synthesis and cell proliferation increased significantly in pcDNA-AHCY transfected B95-8 cells. A modulated expression of surface markers CD11a, CD19, CD23, CD30, CD40, CD44, CD54, and CD58 has not been detected.
