It is demonstrated that extensive monitoring of key parameters (i.e. biomass and product formation) in microscale cultivations enables a profound understanding and hence optimization of recombinant expression systems and high-throughput cultivation processes. New concepts to deal with issues of different growth kinetics of cultures in microtiter plates are elaborated and tested. Furthermore, a novel high-throughput experimentation platform for microscale cultivations is established by combining a pipetting robot with the BioLector, resulting in the Robo-Lector platform. Ultimately, the Robo-Lector can contribute to the envisioned paradigm shift in bioprocess development.
