Einfluss eines kontrollierten Zinkmangels auf die Proteinexpression im Heilungsprozess intestinaler Anastomosen der Ratte

dc:descriptionAlthough intestinal operations are routine surgery, insufficiency is still the most life-threatening complication. That is why research on intestinal wound healing is still of particular importance. Besides operation technique and experience of the surgeon influences of molecular factors are considered to be important for a successful wound healing. The change of the extracellular matrix is one of the-se factors. Prevention of disturbances of wound healing requires a fundamental knowledge of molecular processes. Precondition for intestinal wound healing is an intact collagen metabolism. This kind of interplay between synthesis and degradation constitutes a functional scar. Its regulation is ruled by zinc-dependent proteases (MMPs), which them-selves are regulated by other factors such as TIMPs. A negative zinc balance can disturb the function of MMPs and suppress wound healing. The influence of zinc on physiologic function of the factors, tested in this work, is not known so far. In this study, the expression of TIMP-1, ß-Catenin, c-myc and Notch-3 are checked concerning changes when zinc deficiency is induced in adult rats. An-other examination evaluates the influence of different operation method (anas-tomosis and colotomy) on protein expression. In summary, the expression of the reviewed factors is influenced little by zinc or the operation method. TIMP-1 is totally zinc-independent, ß-Catenin and c-myc are to a large extent zinc-independent. Notch-3 is influenced to some extent due to significant higher scores in zinc deficiency. The operation method is most important for c-myc, which has a higher expression in colotomies. This series of experiments excludes a relevant influence of zinc deficiency and operation method on the here examined factors. The one-phase evaluation of the expression cluster should be considered as a limitation, because it cannot display the different phases of wound healing.
