Therapy treatment options, especially with regard to the non-exudative dry form of macular degeneration, are limited. Some recent studies indicate that choroidal disturbed circulation contributes to macular degeneration. This is primarily caused by atherosclerotic narrowed vessels. In numerous clinic studies it could be shown, that rheophorese treatment can reduce plasma viscosity as well as the tendency towards erythrocyte aggregation. This leads to improvements in choroidal blood circulation and therefore to visus improvements too. Aim of this thesis was to establish an animal-model to analyse hemodynamic and micro-circulatoric properties of the choroidal and retina vascular system with a standard laboratory specimen animal (\"Sprague-Dawley rat\"). The development of this model was necessary for a better understanding of the pathogenesis and for developing therapeutical treatments for the age-related macular degeneration. Therefore the selective ocular blood perfusion was performed on an rats anatomical intact eye using a micro-catheter with erythrocytes suspension having defined rheological properties. By means of fluorescence angiography the perfusion rates had been intravitally recorded using microscope techniques. The quantitative analysis of the choroidal micro-circulation at different perfusion and flowing conditions led to the percentage of homogeneous perfused choroidal regions. Significant different perfusion rates and inhomogeneities dependent on important physiological parameters such as plasma viscosity, aggregation tendency of erythrocytes and vascular width. The results support the expectation that a \"rheopheresis therapy\" can improve the flowing ability and hence the choroidal perfusion crucially, especially in the presence of a pathological initial situation (which often exists in the elderly). Even considering current pathological-genetic approaches to macular degeneration which are focussed on inflammatorial and immunological aspects, the above mentioned results are quite relevant. A disturbed circulation in a constricted vascular system can serve as a promoter for infection reaction. For other diseases too, e.g. diabetic retinopathy, in which a reduced and inhomogeneous perfusion rate by pathologically narrowed vessels can be reported, a therapeutic option based on rheophorese treatment could be considered as a useful therapy.","abstract_html":"The age-related macular degeneration (AMD) is the most common disease in the western civilisation leading to blindness.
