Maus-Herpesvirus-68-(MHV-68)-Infektion : Modell für die Epstein-Barr Virus-Infektion des Menschen

dc:descriptionEpstein-Barr virus (EBV) infection in children may cause severe central nervous system infection with neurological complications like epilepsia and enzephalitis. The well-established model of the EBV-related mouse herpes virus 68 (MHV 68), was not applicable for infections of the central nervous system (CNS) since infections in a natural way were not possible. The aim of this work was finding a strategy which allowed a MHV-68 infection of the CNS. In addition there was to develop a MHV-68-permissive cell line which is suitable for in vitro investigations. The following results were achieved:- infection of the CNS succeeded after intranasal application of MHV-68- detection of viral antigens in the CNS and other organs succeeded by means of immune fluorescence- the virus was succesfully grown from tissue- a suitable dose for intranasal infection was determined- the virus was localised in the CNS- detection of viral m-RNA in tissues was possible, as an expression of replication activity- infectious virus particles were detected in various tissues- a constantly growing, MHV-68 positive B-cell line was developedThe known MHV-68 model has succesfully been extended and can now be used for studies in infections of the CNS. The path of infection and the preferential localisation of the virus in the CNS were shown. These results can be used to examine the pathogenetic effect of the virus for cells and structures of the CNS further.
