Inhibition der intraperitonealen Adhäsion humaner Magenkarzinomzellen in Nacktmäusen
Abstract
dc:descriptionThe inhibition of adhesion of free floating tumorcells to the „mesothel“ is a primary step in the prevention of peritoneal carcinomatosis. In several studies of our group could be demonstrated that phospholipids inhibit the adhesion of tumorcells in vitro and vivo. In this study 100 mice of the “tribe” BALB/c nu/nu have been operated. The experiment consisted of 5 groups with in each 20 animals. Each of the five groups again had 10 animals that were treated with phospholipids and 10 animals that were treated with NaCl solution. Within the laparotomy following concentrations were applicated to the peritoneum: 10.000, 50.000, 100.000, 250.000 and 500.000 cells. 10 animals of each concentration were treated with 150 mg/kg KG phospholipids solution und the other 10 with 0,9% NaCl-solution. The applicated volume was constant in each group. Intraoperativ has been also placed a peritoneal lesion to the caecum. The animals have been put to death after 30 days und examined for the tumorexpanse, tumorvolume, localisation of the tumor und impact on the damaged “mesothel”. The statistic analysis has been calculated with multifactorial ANOVA´s and T-Tests. Phospholipids show in all examined parameters a better result compared with the NaCl solution. With phospholipids treated mice did have significantly less tumorexpanse (p < 0,001) and tumorvolume (p < 0,001). Besides a significantly smaller PCI (p < 0,001) and a smaller amount of tumorspred into the caecum compared to the controllgroups. Sideeffects as inflammation, allergic reaction, woundhealingdisorders and early deaths have not been noticed.
