The Use of Morphogenic Transcription Factors to Enhance Plant Regeneration
… removed) for transient expression and subsequent regeneration of non-GMO, edited plants. However, efficient regeneration of plants from protoplasts is a bottleneck in the implementation of this technique. To create a universal method for protoplast regeneration, there first needs to be a baseline …

Onion (Allium cepa L.) seed aging and plant regeneration in vitro
Onion seeds had cracks, and fungal infection was found over and inside the hilum and seed cracks. During storage dehydration and fungal infection may lead to membrane damage. Aged seeds had a high electrolyte leakage and encountered hydration stress which reduced seed germination and seedling vigor.

Efficient production of doubled haploid plants through antimicrotubule agent treatment of anther-derived maize callus and the RFLP analysis of the maize anther culture response
… of maize anther-derived haploid callus before plant regeneration. Without treatment with these agents, all the plants regenerated from the callus were haploid. More than 50% of the plants regenerated from the callus treated with 0.625 or 1.25 uM colchicine for 1 to 3 days, 5 to 10 uM APM or 10 …

Biolistic-mediated transformation of Eragrostis Curvula with the HSP 12 gene
… is a desiccation sensitive monocotyledonous plant and an economically important forage grass in southern Africa. This species has a potential to be improved for drought and salt resistance among other important agronomic traits. In this study, E. curvula was used as a model system to explore …

Expanding the genetic variability of flatpea using tissue culture, mutagenesis, and intercrossing techniques
… of interest, three approaches were utilized: (1) regeneration of flatpea plants from tissue culture to produce potential somaclonal variants, (2) seed irradiation and screening of potentially mutated progeny, and (3) intercrossing among flatpea accessions. Low-frequency whole plant regeneration of …

Genetic engineering for pest resistance in potato (Solanum tuberosum L.)
… way for targeting genes of interest into the plant genome consists of Agrobacterium-mediated gene transfer. This thesis studied Agrobacterium-mediated transformation to transform two potato cultivars with an avidin gene anticipated to confer insecticidal activity against certain lepidopteran …

Urban Restoration Ecology: Investigating environmental change, ecological function, and enrichment planting
… forests, it is unclear how dynamics change after planting and over the long-term, and hence what best management practices should be used in the decades following initial plantings. This thesis addresses these knowledge gaps through three separate studies on restored urban forests. The first study …

Development of genome editing in potato and tobacco
… et al., 2010). Studies on Arabidopsis and potato plants have shown that Glucan Water Dikinase 1 (GWD1) plays a crucial role in the first step of starch degradation (Kozlov et al., 2007; Wickramasinghe et al., 2009; Hejazi et al., 2014; Pirone et al., 2017; Ohnuma et al., 2023). Silencing of the …

Development and application of novel genetic transformation technologies in maize (Zea mays L.)
Plant genetic engineering approaches are of pivotal importance to both basic and applied research. However, rapid commercialization of genetically engineered crops, especially maize, raises several ecological and environmental concerns largely related to transgene flow via pollination. In most …

Genetic Engineering of 2,4-D Resistance in Wine Grapes
… 2,4-D easily drifts and severely affects grape plantings around areas where it is used. Since grapes have no known genetic resistance to this herbicide, breeding to incorporate resistance in commercial cultivars cannot be done. Thus genetic engineering has to be employed to produce adaptable …

Soil Microbial Community Dynamics In Florida Scrub Ecosystem
… the natural fire cycle. The effects of fire over plant regeneration and fauna habitat utilization of the scrub have been well characterized in previous research. In the present paper the objective is to characterize the short- and long-term fire effects on the soil microbial community. Fire …

Effects of Elevated Atmospheric CO2 on Root Dynamics, Biomass and Architecture in a Scrub-Oak Ecosystem at Kennedy Space Center, Florida
<p>A major gap in whole-plant ecology lies with our understanding of root system growth, function and distribution. Large belowground structures, in addition to fine roots, are of particular interest because of their role in carbon sequestration. Non-destructive methods, including …

Germination thresholds of the Mixed-grass Prairie species as affected by global climate change: A FACE study.
… of global climate change on seed germination and plant regeneration have been reported in many species. However, there are no consistent trends in how seed quality and germination are affected by these conditions. Seeds of four native, one invasive, and two pairs of native/invasive species were …

Inheritance of protoplast culturability and improvement in pollen development by protoplast manipulation in solanum
… protoplast culturability and regenerability of a plant. The ability of callus formation and plant regeneration from protoplasts varies among plants. To understand the genetic basis for this variation, the mode of inheritance for protoplast culturability, defined as the ability to develop calli …

PROPAGATION OF PLANTS BY SOMATIC EMBRYOGENESIS
The in vitro culture of plants offers many advantages over conventional propagation but its wider commercial application is largely limited by high labour costs. These would be reduced by utilising the phenomenon of somatic embryogenesis for plant regeneration (Jones, 1984, Biochem. Soc. Symp. 98: …
