Análisis de polimorfismos de genes implicados en la regulación del endotelio y la inflamación en pacientes con insuficiencia cardíaca y su relación con el tipo de disfunción ventricular predominante

Abstract

[EN] Heart failure is determined by genetic and environmental factors. Endothelial system and the inflammatory response are essential in vascular remodeling in heart failure. Polymorphisms of genes encoding endothelin-1 (ET-1), the endothelial nitric oxide synthase (NOS), interleukin-6 (IL-6) and cyclooxygenase-2 (COX2) could be related to ventricular remodeling in patients with heart failure. Our objective was to investigate the relationship between polymorphisms of these genes: Lys198Asn (ET-1), Glu298Asp (NOS3), 765G> C (COX2) and-174G> C (IL6) and the development of ventricular remodeling in patients with renal heart. We conducted a case-control study which included 190 patients with heart failure, 105 dilated left ventricle and 85 with non-dilated left ventricle. DNA was extracted from patients using standard protocols. The NOS3 gene polymorphism selected was analyzed using restriction endonucleases. The rest of polymorphisms were analyzed by using Taqman probes aplied Biosystems thermocycler 7000. The T allele of the polymorphism Lys198Asn of endothelin-1 was more prevalent in a statistically significant in patients with left ventricular dilatation (40% vs 21.2%, p <0.000). No differences were found in the other polymorphisms studied in relation to left ventricular dilation in patients with heart failure.
