Investigating the Role of Adaptor Protein ShcD in the Oligodendrocyte Lineage

dc:description.abstractOligodendrocytes (OLs) are the myelin producing cells of the central nervous system, which are required for the coordination of neuronal signaling. Adaptor protein ShcD is highly expressed in oligodendrocyte progenitor cells (OPCs), with a gradual reduction across differentiation to mature OLs. Utilizing ShcD wildtype (WT) and ShcD knockout (KO) mice, we investigated the role of ShcD within this lineage. We probed the expression of OL lineage markers in mouse brain tissue and found a reduction in mature OL markers in rostral corpus callosum containing regions. In addition, we generated ShcD WT and ShcD KO primary OPCs that are capable of being differentiated into mature OLs. ShcD KO OPCs showed a reduced capacity for migration and reduced complexity early in differentiation compared to their WT counterparts. These findings indicate a role for ShcD in OPC migration and differentiation, suggesting its involvement in developmental myelination and remyelination following injury.
