Abstract
dc:descriptionThe toxic action of hyperoxy in tissues is due to the formation of oxygen intermediants (free radicals and toxic metabolites). The purpose of this study is, to search the effects of normobaric hyperoxy on the structure of the hepatic cell and on SGOT and SGPT values. Protection of the hepatic cell with administration of vitamin E, a substance with noted antioxidantive action, was also studied. Seven (7) groups of wistar laboratory rats consisting of ten (10) animals each were studied. Four (4) of the groups were exposed to normobaric hyperoxy (100% Ο₂) for 24, 48, 60 and 72 hours respectively. The other three (3) groups were exposed to hyperoxy 48, 60 and 72 hours respectively after having been fed 15mg/kg BW/24H of vitamin E for fourteen days with their regular feed. The rats were killed and endocardiac blood samples were taken to determine aminotransferase values using phasmatophotometric method. Hepatic tissue sections were studied with the electronic microscope. Damaged mitochondria, smooth endoplasmic reticulum and damaged cytoplasm were observed in the rat groups, which had been exposed to 48 hours of hyperoxy without previous administration of vitamin E. Damages became more intense with basic intercellular damage at 60 hours of hyperoxy. Also SGOT and SGPT values were increased (240 and 82, 62 units respectively after 48 hours of hyperoxy and 700 and 287,25 units respectively after 60 hours). In the groups, which received vitamin Ε, nο significant damages of hepatic cells were observed at 48 hours and 60 hours of hyperoxy. Aminotransferases in these groups were increased after of 48 hours of hyperoxy (SGOT 206,82 SGPT 68,62), but at 60 hours of hyperoxy, normal SGPT values (22,25) and slightly increased SGOT values (73,25) were observed. In comparison aminotransferase values between groups that were given vitamin E and those not given, varied significantly after 60 hours of hyperoxy, although at 48 hours no significant differences appeared. The rat groups exposed to 72 hours of hyperoxy, were excepted from the study, because arterial blood gas oxygen values were found to be less than 100 mm/Hg. From the results of this study it is evident, that exposure to normabaric hyperoxy produces, damages to hepatic cells and increases aminotransferase values. Administration of vitamin E may protect the hepatic cell, due to inhibition of hyperoxidation of lipids from the toxic oxygen intermediants. However it does not prevent lung tissue damage.
