ΥΠΟΔΟΧΕΙΣ ΑΔΕΝΟΣΙΝΗΣ. ΕΠΙΔΡΑΣΗ ΝΟΥΚΛΕΟΤΙΔΙΩΝ ΤΗΣ ΓΟΥΑΝΟΣΙΝΗΣ ΚΑΙ ΔΙΣΘΕΝΩΝ ΜΕΤΑΛΛΩΝ
Abstract
dc:descriptionINVESTIGATION OF THE EFFECT OF GUANOSINE NUCLEOTIDES AND DEVALENT CATIONS ON ADENOSINE RECEPTORS. THE BIOLOGICAL SYSTEM THAT HAS BEEN USED WAS RAT AND MOUSE MEMBRANE HOMOGENATES. WE USED BINDING ASSAY METHODS TO STUDY THE BINDING CHARACTERISTICS OF STABLE TRITIADED ADENOSINE ANALOGS THAT HAVE BEEN PROPOSED AS A1 AND A2 SPECIFIC AGONISTS. GTP AND ITS STABLE ANALOG GPP(NH)P CAUSE DECREASE IN THE NUMBER OF BINDING SITES (BMAX) OF 3H-CHA AND 3H-PIA. IN CONTRAST DEVALENT CATIONS CAUSED A SMALL INCREASE IN BMAX OF THE SAME ANALOGS. IN THE PRESENCE OF THE TWO FACTORS BINDING CHARACTERISTICS OF ADENOSINE ANALOGS WERE THE SAME WITH CONTROL. FROM KINETIC STUDIES WITH 3H-CHA AND GPP(NH)P IT WAS FOUND OUT THAT ADENOSINE RECEPTORS EXISTS IN THREE POSSIBLE CONFORMATIONS. THE RELATIVE RATIO OF THESE CONFORMATIONS DEPENDS ON EXTERNAL CONDITIONS. THE THREE CONFORMATIONS ARE: A) CONFORMATION IN WHICH THE RECEPTOR HAS NO ADENOSINE AGONIST BINDING, B) CONFORMATION OF FAST BINDING, C) CONFORMATION OF SLOW BINDING.
