Abstract

dc:description.abstract

The Nuclear Magnetic Resonance spectroscopy is an established technique in the structural elucidation of chemical compounds. This technique is based on the property of the spin isotopes in the presence of an applied magnetic field to assume two different spin states, one aligned with the field - low - energy - and the other in the opposite direction to the applied field - high - energy. Albeit these spin states are almost equally populated , there is a small exces s of spins that assume the low energy state and this excess is responsible for the observation of the phenomenon of resonance. Resonance is achieved when the sample is irradiated and absorbs radiofrequency pulse with energy matching the difference of the t wo spin states, promoting thus the nuclei to higher energy spin states. This study aimed to conduct a literature review on the applicability of Nuclear Magnetic Resonance Spectroscopy of Hydrogen with quantitative purpose in the pharmaceutical and related. The 1 H - q NMR assumes that a given signal intensity is directly pro portional to the number of nuclei that generated it, thereby allowing the concentration of an analyte X to be determined by the integrated area using the signal of a resonance standard for c omparison . The 1 H - qNMR although still underused nationally, has a number of advantages over conventional techniques in quantitative analysis, including: in 1 H - qNMR reference standard need not to be structurally related to the analyte of interest, sample pr eparation is relatively simple, it is possible to identify and quantify various substances simultaneously in one spectrum and is a non - destructive method. This range of benefits has aroused interest in many researchers and today the technique has been empl oyed in the purity analysis, evaluation of industrialized product tampering metabolomics , regulatory toxicology etc. The study of possible combinations and changes of parameters to optimize the technique for quantita tive purposes is a perspective and the t echnique has already matured internationally .
