Desmoteplase zur lokalen Fibrinolyse intrazerebraler Blutungen in einem Großtiermodell
Abstract
dc:descriptionRecombinant Desmoteplase was used for lysis of intracerebral hemorrhages in a well-established large-scale animal model. The fibrinolytic potential of the plasmonogen activator, as well as the effect on the delayed edema was evaluated which is most prominent three to five days after ictus. We observed a significant reduction in hematoma volume by DSPA-lysis (75%), comparable to rtPA-lysis. The delayed edema was not aggravated significantly by DSPA whereas an enlargement of perihematomal edema (413%) was observed in rtPA-treated pigs, comparable to the results of a previous study by Thiex et al. Histological examination of HE-stained brain slices show an increased inflammatory reaction of the perifocal area in tPA-treated animals but not in brains of DSPA-treated pigs. DSPA is a fibrinolytic that could potentially improve the treatment of spontaneous intracerebral hemorrhages if applied locally by minimally-invasive technique.
