T-Zell-unabhängige MHC-II-Signaltransduktion durch Superantigene

For a couple of years MHC-II complexes are known for many more assignments than antigen presentation. In this thesis the quite unknown mechanism of superantigen-induced T cell independent MHC-II-signaltransduction is investigated. It could be demonstrated that the superantigen-induced MHC-II-signaltransduction depends on the existence of lipid rafts. Another result was the finding that the proteinkinase C (PKC) pathway is involved in signaltransduction. Cell aggregation induced by the used anti-MHC-II-antibodies was not significantly decreased by blocking PKC. Furthermore the experiments show that there is no difference in cell aggregation by superantigens with homogenic or heteogenic binding sites for MHC-II, and that the superantigen-induced MHC-II-signaltransduction does not induce apoptosis.
