Die MMP9- und OPG-Expression von dermalen Fibroblasten und mesenchymalen Stammzellen nach Stimulation durch Myelom- und Hodgkinzelllinien
Abstract
dc:descriptionTo understand the pathogenesis of multiple myeloma it is fundamental to know the process of osteolysis. This thesis was supposed to show the possibilities of humeral stimulation of mesenchymal cells by different myeloma cell lines (U266, IM9 and RPMI8226), in order to comprehend the role which mesenchymal cells play during the development of osteolytic focuses in the multiple myeloma. The theory of reduced expression of the osteoprotective factor OPG by fibroblasts could be demonstrated after stimulation with conditioned mediums by the celllines U266 and IM9. Some matrix metalloproteinases seem to play an important role in the domain of angiogenesis of carcinomas, especially MMP9. The attempts of fibroblaststimulation by the hodgkin-cellline HDLM2 showed a manifest increase of MMP9-expression. Both experiments proved the importance of mesenchymal cells in tumor pathogenesis in cases of multiple myeloma and hogdkin lymphoma.
