Tierexperimentelle Untersuchungen zur Adipogenese in Gel-Trägern als Weichgewebsersatz

The goal of this study was to develop a tissue engineered construct on the basis of biocompatible materials and autologous preadipocytes. Three different biomaterials (matrigel, collagen g , HYADD-3) were examined regarding their potency to induce adipose tissue. All three gels were implanted in a standardised chamber made out of acetal polymer into immune deficient naked rats. In order to assess the angiogenetic potency of the different gel-based carriers two different types of vascularisation (axial and randomised) were created. Axial vascularisation was produced by transposition of femoral artery and vein into the chamber. Randomised vascularisation was achieved by direct contact to subcutaneous tissue. Neotisssue formation in Matrigel implants was unspecific for adipose tissue formation. After forty days of implantation neoadipogenesis could be noted in the axially vascularized implants. Collagen and hyaluronic acid implants produced similar results. Only axially vascularized implants revealed an adequate angiogenetic potential to induce neotiissue formation. HYADD-3 gel which is beased on the amide of hyaluronic acid was able to produce the highest amount of adipose tissue and had the longest in vivo stability. Collagen gel had in comparison a shorter in vivo stability and lower amounts of adipose tissue. Microscopic evaluation of the biopsies revealed tissue that mainly consisted of fibrocytes and adipocytes. This study uses a new method to evaluate tissue engineering constructs in vivo. It also brings up for the first time a new possibility for adipose tissue augmentation based on an autologous preadipocytes – hyaluronic acid gel model.
