Collagen sponges are used in medicine because of their good biological and physicochemical properties and suitability for drug delivery, reconstructive skin surgery, and replacing lost tissue in subjects with burn injuries. A number of collagen matrices are currently in clinical use and available commercially. This study was conducted to investigate a new collagen matrix intended for medical use as a hemostyptic and for dressing wounds. The results constitute an important contribution in obtaining certification for the collagen sponge as a medical device. The collagen sponges investigated in this paper were manufactured on a large scale from a porous collagen sheet comprising several layers. One of the main intentions of this study was the biological and physicochemical characterization of matrices from defined layers, especially from the upper, middle and lower layer. The research work involved in vitro cytotoxicity studies on the basis of material extract tests with fibroblast cell line L929 in accordance with ISO 10993-5. The effect on cell growth of Gamma-sterilized matrices and that of plasma-sterilized sponges exposed to different degassing times was studied with L929 cells, as was the concentration-dependent effect of insulin (as a drug potentially suitable for delivery via collagen sponge). In addition, the microstructure (pore surface and diameter) was analyzed by ESEM and the degradation behavior of the sponges was studied in various media (bidistilled water, HUVEC and GM7373 medium). Another focus of the studies was the in vitro growth behavior of cells (L929 fibroblasts, GM7373-endothelial cells, PC12-neuroectodermal cells) on non-modified and EDC/NHS-crosslinked collagen sponges in association with various colonization and cultivation methods. The tolerability and absorption of collagenous Gamma-sterilized original matrices were studied in a chick embryo chorioallantoic membrane model in preparation for future in vivo studies in porcine models.
