Intraperitoneale Reduktion der Tumorzelladhäsion mit Phospholipiden : eine tierexperimentelle Studie

dc:descriptionThe formation of secondary peritoneal carcinosis is the outcome of intraoperative deportment of tumor cells in the peritoneal cavity. This study deals with the pathomechanism and especially with the adhesion of the colonic tumor cells on the extracellular matrix within the scope of an animal experiment. The positive results in the postoperative prevention of adhesion explain the use of the phospholipids in this study. The formation of adhesions between peritoneum and other tissues or organs predominantly takes place in the lesions of the extracellular matrix. The intraperitoneal application of the phospholipids causes a membranoid protective coating during the wound healing of the lesions. This happens in the case of tumor cell nidation, too. So the intraperitoneal used phospholipids also in this case serve as prevention of adhesion. The study is orientate by the following questions: 1) Is it possible to reduce the adhesion of the tumor cells with intraperitoneal application of phospholipids? 2) Does this effect leads to a reduce of peritoneal carcinosis? 3) Is it possible to improve the survival rate with intraperitoneal application of phospholipids? 4) Does the influence of the phospholipids depends on the concentration? A total of 90 female BD-IX rats were divided into three big groups with 30 animals with treatments of phospholipids at 75mg/kg or 150mg/kg bodywight or sodium chloride at 0,9% in the control group. Every animal was a definite number of colonic tumor cells (DHD-K12-Trb) implanted. After 30 days, the extent of peritoneal carcinosis was determined by measuring the tumor volume, the area of tumor attachmend and the Peritonea lCancer Index. The Peritoneal Cancer Index is a score built out of different values to describe the extension of the peritoneal carcinosis. Over a 90-day observation period, the survival rate was analyzed. The results show significant difference between the control group and the phospholipids group in view of the tumor volume, the area of tumor attachmend and the Peritonea lCancer Index. In view of the survival rate we get an improvement but no significant difference. We find out a significant difference between the 75mg- and the 150mg-phospholipidconcentration as well. In summary it may be said that the intraperitoneal application of the phospholipids causes a reduce of the adhesion of tumor cells and as aresult leads to a reduce of peritoneal carcinosis. The study shows that the effect of prevention of adhesion depends on the concentration of the phospholipids. There are signs of improvement in the survival rate of the animals. Additional examinations have to show if the discribed effects may be improved by a modification of the therapy.
