The present doctoral thesis is divided into two parts. In the first part a further method for the clonality analysis X-chromosomal-linked genes – besides the often used androgen-receptor-assay – was established on probes of uterine leiomyomata. The chosen gene locus was that of the MAOA gene, which has already been useful for this PCR-analysis at lymphomas and hematopoietic cells of heterocygous women. This MAOA-assay was shown to be well suited for the clonality analysis of fresh frozen tissue. The thesis lists the pros and cons in comparison to the androgen-receptor-assay and to the bisulfit-modification, which is also used for clonality analysis in the literature. In the second part this clonality analysis was applied to the clinical picture of the "benign metastasizing leiomyoma" ("BML") for to clear up its pathogenesis. What is special about it is that the BML is a histological benign tumour, which though shows a clinical malign growth behaviour with metastases. With the clonality analysis it was possible to prove a monoclonal process on all of the probes of one patient, who was suffered from this disease. Therefore one of the three hypotheses about the pathogenesis of the BML - that one, which favours the presence of a multifocal (that means polyclonal) process - could be exposed as to be quite unlikely or even defeated. Not yet finally proved is the question about the benignancy or malignancy of this disease. That schould be the aim of future analyses.
