Filmbasierte automatische Detektion und fraktale Analyse des Mikrozirkulationsnetzwerkes der Chorioallantois-Membran des Hühnereies

dc:descriptionThe microvascular network of the chorioallantoic membrane (CAM) is frequently used as a model to investigate angiogenesis. Up to now, the quantification of the vessel system has been unsatisfactory because it was performed by a subjective classification without regard to the capillaries. For the detection of the transparent capillaries the movement of the erythrocytes was used in a non-invasive process. To this end, the erythrocyte flow was recorded in a digital film sequence. To detect the capillaries the movement of the membrane was separated from the erythrocyte movement. The capillaries were separated from the background by analyzing a histogram. The result is a binary image of the microvascular network. A wave-like algorithm was established to extract the interesting parameters of the vessel system . The influence of noise can be reduced by this algorithm. Also the numbers of bifurcations, the cross section of the vessels and the length between two bifurcations can be evaluated. The influence of biomaterials, like prosthetic meshes for hernia repair, was quantified by the fractal dimension. It was shown that the fractal dimension is a function of the distance to the biomaterial. With this new method the microvascular network of the CAM can be evaluated automatically. It can also be used for all angiogenetic tests on the CAM, such as the investigation of tumor tissue.
