Untersuchungen zur Wirkung des Hepatocyte Growth Factor (HGF) auf Humane Omentum Majus Zellen

dc:descriptionFrictionless movement plays a pivotal role in the function of intraabdominal organs. During inflammatory or traumatic processes of these organs, the peritoneum reacts with the secretion of a fibrin-rich exsudate. Imbalance of pro- and antifibrinolytic enzymes may lead to permanent adhesion of the serous layers, which could for example provoke an ileus. Elevated levels of hepatocyte growth factor (HGF) were found in serum analyses of patients who had undergone liver surgery. In order to get insight into peritoneal wound healing, which occurs during liver regeneration, the presented study investigated on the influence of HGF on cultured human mesothelial cells (HOMC), according to their expression of the c-met receptor, the secretion of the fibrinolytic enzyme t-PA (tissue-plasminogen-activator) and the secretion of the antifibrinolytic enzymes PAI-1 and PAI-2 (plasminogen-activator-inhibitor 1 and 2). Furthermore the influences on proliferation and migration were studied. With the use of immunehistochemical techniques, the c-met receptor could be detected on mesothelial cells. Stimulation of HOMC with HGF in a concentration of 0-100 ng/ml led to an elevated secretion of t-PA and PAI-1. Slightly increased secretion of PAI-2 was observed, only when mesothelial cells were stimulated with HGF in a concentration of 100 ng/ml. To present an in vitro model for peritoneal wound healing, mesothelial cells were scratched out of a confluent cell monolayer using the tip of a pipette. The coverage of these defects was measured after eight hours. A significant faster coverage of the defects was detected under stimulation with HGF. This effect is based on an elevated migration and not on an increased proliferation
