Synthese des UDCA-7[beta]-N-Acetyl-D-glucosaminids und Stoffwechsel der Isoursodeoxycholsäure (IsoUDCA) beim Menschen

dc:descriptionThe aim of the first part of this thesis was the non-enzymatic synthesis of 7ß-N-acetyglucosaminides of UDCA. This conjugate should serve for in vitro investigations of the mechanisms of UDCA. The synthesis was very complex and difficult, and the yield was quite small. The aim of the second part of the study was to examine the metabolism of isoursodeoxycholic acid (isoUDCA) in humans. IsoUDCA was synthesized with >99% purity> It was administered orally to six healthy males for 1 week (3x250 mg daily). Bile acid were extracted from duodenal bile, serum, and 24-h urine samples collected before and at the end of the study period, separated into distinct conjugates, and subsequently analysed by gas chromatography mass spectrometry as well as fast atom bombardment mass spectrometry. This study demonstrates good tolerance and significant intestinal absorption of orally administered isoUDCA. IsoUDCA is extensively isomerised, probably both by intestinal and hepatic enzymes to yield UDCA, which becomes the major biliary compound.
